The plot centres on the story of Smurfette, and how she came to be the only girl in Smurf village. Singer Katy Perry voices the female Smurf, providing her first lead role in a film. However, Perry was, according to Kerner, chosen "completely anonymously from a range of voices we were asked to listen to". Perry insists she is not interested in furthering her acting career "as there's too many singers out there who think they can act, and too many actors who think they can sing - it doesn't always work that way".
Surprisingly, it is not Perry who provides the soundtrack for the movie. If it all sounds light years away from the innocence of Father Abraham's hit Smurf Song, Kerner insists the family of the creator, Peyo, who died in , approves of any changes.
But they appreciate that we have to move with the times in other ways, to make it relevant to our audience. However, when it comes to filming the Smurfs, Neil Patrick Harris adds, it's still done using the traditional methods of storyboarding, followed by markers on set to denote where a Smurf would be standing in a scene.
The animation is added in later. It helps that the markers now have little LED lights on them all around, representing which Smurf is standing where. They light up according to which one is talking, so you know where to look.
At the same time, Smurfette tries to escape, but Vexy and her partner Hackus catch up to her, with Vexy convincing Smurfette that she isn't alone, and together the Naughties bond with Smurfette and have fun. Upon her return to Gargamel's hotel suite with the Naughties, Gargamel presents her with a gift, her very own magic wand, as a feigned act of kindness, claiming that he was Smurfette's father all along and that Papa Smurf had no interest in her.
Yet Smurfette is still reluctant to give Gargamel what he wanted until she sees that the Naughties are dying due to a lack of Smurf essence. Realizing that the only way to save them is to give Gargamel the secret formula, Smurfette quickly writes it down and Gargamel mixes it up to turn the Naughties into real Smurfs. But immediately after they become Smurfs, the evil wizard puts them into his Smurfalator so he can carry out the rest of his plan.
Meanwhile, Patrick, Victor, and the Smurfs work together to rescue Smurfette from Gargamel when the Smurfs get captured and put into the Smurfalator along with Smurfette, Vexy, and Hackus, using it to power the evil wizard's bigger dragon wand. Patrick and Victor arrive in time to destroy the Smurfalator, causing the Smurf-creating formula to be destroyed in a fire, but not before Gargamel gained enough power to use his new wand against the Smurfs and their allies.
Vexy and Hackus teamed up with the Smurfs against their former master and used the new wand to blast him away, far up into the stratosphere.
With Gargamel no longer a threat and with two new Smurfs to add to the family, the five time-traveling Smurfs make their goodbyes to the Winslows and return home to celebrate Smurfette's birthday.
